Mutations in the patched gene have been found to underlie the basal cell nevus syndrome in which patients develop innumerable basal cell carcinomas, as well as odontogenic jar keratocysts, craniofacial and musculoskeletal abnormalities, palmar pits, and a variety of other tumors including medulloblastomas.
For example, scientists recently discovered that mutations in the human version of a gene that controls fruit fly growth and development are the likely cause of both the basal cell nevus syndrome, a rare inherited disorder, and sporadic basal cell carcinoma of the skin, the most common human cancer.
Discovery of the new gene is "incredibly significant in terms of understanding the pathogenesis of basal cell nevus syndrome and basal cell carcinomas in general," comments Bert Vogelstein of the Johns Hopkins Medical Institutions in Baltimore.
