restriction enzyme

Bacterial enzyme that breaks a chain of DNA into two pieces at a specific point; used in genetic engineering. The point along the DNA chain at which the enzyme can work is restricted to places where a specific sequence of base pairs occurs. Different restriction enzymes will break a DNA chain at different points. The overlap between the fragments is used in determining the sequence of base pairs in the DNA chain.
