Typically a burette is filled with an acid of unknown concentration which is slowly (drop by drop) added to a known volume of an alkali of a known concentration, mixed with an indicator (such as phenolphthalein). The volume of acid need to neutralize the alkali in the flask can be used to calculate the concentration of the acid.
