Isoelectric focusing showed that AX476 produced a [beta]-lactamase with an isoelectric point of 7. Separation can be done simply at the whole-protein level using two-dimensional gel electrophoresis; this technique separates molecules by molecular weight and isoelectric point (the pH at which a molecule carries no net electrical charge). The mixed-mode sorbents can distinguish proteins that have similar or very close isoelectric points, a separation that cannot be achieved with a single ion exchange step. |
